UNC93B is a member of a novel hUNC93B-related gene family and encodes a 12-membrane-spanning protein related to unc-93 of Caenorhabditis elegans. The protein is involved in the regulation or coordination of muscle contraction in the worm. The protein is also necessary for TLR signaling and for the presentation and cross-presentation of exogenous antigens. The protein specifically interacts with TLR3, 7, 9, and 13 and introduction of point mutation H412R in UNC93B abolishes these interactions. Mutation in the missense allele of UNC93B, 3d disrupts exogenous antigen presentation and signaling via TLR. The pathological role of the protein has been identified in susceptibility to herpes simplex encephalitis (HSE). In human the protein is expressed in heart and at lower levels in kidney. In mouse the gene is localized in the chromosomal region 19 A.
