This antibody is specific for the VGLUT1 protein. On western blots, it identifies the target band at ~55 kDa. Reactivity has been confirmed with mouse brain homogenates by western blotting, with mouse hippocampus frozen sections by immunohistochemistry, and in immunofluorescence analysis with human glutamate neurons derived from iPSCs.

VGLUT1 (Vesicular Glutamate Transporter 1, BNPI, SLC17A7) protein is a vesicle-bound, sodium-dependent inorganic phosphate transporter that functions as a vesicular glutamate transporter and may be involved in neuronal sodium transport. Vesicular glutamate transporters VGLUT1 and VGLUT2 show a mutually exclusive distribution in the adult brain that suggests specialization for synapses with different properties of release. Consistent with this distribution, inactivation of the VGLUT1 gene silenced a subset of excitatory neurons in the adult brain. The targeted deletion of VGLUT1 in mice caused lethality in the third postnatal week, a time when there is a developmental switch from VGLUT2 to VGLUT1 expression. Glutamatergic neurotransmission was drastically reduced in neurons from VGLUT1-deficient mice, with a specific reduction in quantal size. The remaining activity correlated with expression of VGLUT2. It indicates that the level of the VGLUTs expressed determines the amount of glutamate that is loaded into vesicles, thereby regulating the efficacy of neurotransmission.
