Description
Normal cardiac morphogenesis requires a number of transcription factors, including Nkx2.5, MEF2C, eHand, and dHand. Both bone morphogenetic protein-(BMP) and Nkx2.5- dependent pathways have been implicated in intrinsic cardiogenesis. Xin, a BMP-induced protein, is expressed in embryonic and adult cardiac and skeletal muscle. It contains an N-terminal DNA binding domain, followed by a region of 16-amino acid repeating units, a nuclear localization signal, and a SH3-binding motif. In adult mice, Xin colocalizes with N-cadherin and connexin-43 in the intercalated discs of the heart, while in neonatal cardiomyocytes, Xin is found at cell-cell contacts and in the nucleus. In chick embryos, incubation with antisense oligonucleotides to cXin causes abnormal cardiac morphogenesis and an alteration in cardiac tube looping. In addition, BMP induces the expression of Nkx2.5, MEF2C, and cXin in chick embryos. This induction of Xin may occur downstream in this pathway, since both Nkx2.5 and MEF2C can act as Xin transactivators. Thus, Xin is thought to be involved in cell-cell junction dynamics and nuclear signaling during BMP- and Nkx2.5-dependent cardiac morphogenesis.
