The BRD9 gene encodes a protein that is a crucial subunit of the SWI/SNF chromatin remodeling complex known as ncBAF (non-canonical BAF complex). As a bromodomain-containing protein, BRD9 acts as an 'epigenetic reader,' selectively recognizing acetyl-lysine marks on histones, which is essential for regulating chromatin structure and gene expression through the recruitment of acetylated transcriptional regulators. It has emerged as a significant player in epigenetic modulation and has been implicated in various cancers, such as cervical and non-small cell lung cancer, due to its over-expression. BRD9 is situated on the 5p arm of chromosome 5 and is a potential target for anticancer therapies, notably in acute myeloid leukemia (AML). Recent studies highlight the development of selective BRD9 inhibitors, indicating its potential for therapeutic targeting.
