Description: This monoclonal antibody 47.7G7.1F2 reacts with human CD126, also known as IL-6 receptor alpha. It associates with the gp130 chain to form the high-affinity receptor for IL-6. Although gp130 is ubiquitously expressed, CD126 is only found on mature T cells, activated B cells, macrophages, hepatocytes, and myeloma cells. Upon binding to IL-6, two heterotrimeric complexes of receptor and ligand associate into a hexamer and are then internalized, initializing Jak/STAT signaling pathways. IL-6 induces the proliferation and survival of T cells, Th2 differentiation, and acute-phase protein production in the liver. A soluble form of CD126 lacking the transmembrane domain is also present and can be generated by cleavage at the cell surface or alternative splicing. Soluble CD126 can bind IL-6 and activate cells expressing only gp130 through trans-presentation. Applications Reported: This 47.7G7.1F2 antibody has been reported for use in flow cytometric analysis. Applications Tested: This 47.7G7.1F2 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 μL (0.06 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.

Interleukin 6 receptor (IL6R), also known as CD126 (Cluster of Differentiation 126), is a type I cytokine receptor. This gene encodes a subunit of the interleukin 6 (IL6) receptor complex. Interleukin 6 is a potent pleiotropic cytokine that regulates cell growth and differentiation and plays an important role in the immune response. The IL6 receptor is a protein complex consisting of this protein and interleukin 6 signal transducer (IL6ST/GP130/IL6-beta), a receptor subunit also shared by many other cytokines. Dysregulated production of IL6 and this receptor are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer. Alternatively spliced transcript variants encoding distinct isoforms have been reported. A pseudogene of this gene is found on chromosome 9.
