Rat Monoclonal Antibody

Product is supplied at 1 mL (100 μL/mL). This antibody recognizes mouse CD200 Receptor-like 3 (CD200R3), a protein of ~38 kDa or ~74 kDa (reduced or non-reduced condition, respectively). Application Notes: Western Blot - Antibody was used in reducing and non-reducing conditions with MC/9 lysates. Under reducing conditions, the resulting band size was 38kDa, and under non-reducing conditions, the resulting band size was 74 kDa. Flow Cytometry - Basophils and mast cells were used. No fixation is needed. Functional Studies - Antibody was used as an agonist in basophil-mediated allergic reactions. A single intravenous administration of 30 μg antibody was used . The biological activity of the antibodyis a drastic reduction in the basophile number in the peripheral blood and spleen, to approximately 20% of the normal level. This product is also useful for mast cell degranulation. Readout was determined by measuring hexosaminidase activity. Immunoprecipitation - mast cells were lysed with 1% NP-40 containing lysis buffer with proteinase inhibitor mixture before incubation with the antibody.

CD200R3 is a dimeric cell surface receptor also known as CD200RLb. CD200R3 contains two Ig-like extracellular domains and as such is a member of the immunoglobulin superfamily of receptors. The intracellular region of CD200R3 associates with the adaptor molecule DAP12, and possibly also DAP10, CD3 zeta and FcR gamma-chain, indicating that it is capable of transducing stimulatory signals. In this respect CD200R3 bears similarity to activatory receptors CD200R2 and CD200R4. In contrast, an inhibitory receptor CD200R (also known as CD200R1) relies on other adaptors. CD200R3 is primarily expressed by basophils and mast cells, cells known to be involved in anti-parasitic immune responses and frequently also allergic reactions. The surface expression of CD200R3 decreases when basophils are activated by ligating their CD16 and CD32. Remarkably, most of this downregulation is observed within the first 30 minutes after activation. Unlike CD200R, CD200R3 does not bind CD200.
