Rat Monoclonal Antibody

Description: This Ba13 monoclonal antibody recognizes mouse CD200R3, a surface receptor expressed by basophils and mast cells. Applications Reported: This Ba13 antibody has been reported for use in flow cytometric analysis. Applications Tested: This Ba13 antibody has been tested by flow cytometric analysis of the MC/9 cell line. This may be used at less than or equal to 0.25 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Light sensitivity: This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. CD200R3 is a dimeric cell surface receptor also known as CD200RLb. CD200R3 contains two Ig-like extracellular domains and as such is a member of the immunoglobulin superfamily of receptors. The intracellular region of CD200R3 associates with the adaptor molecule DAP12, and possibly also DAP10, CD3 zeta and FcR gamma-chain, indicating that it is capable of transducing stimulatory signals. In this respect CD200R3 bears similarity to activatory receptors CD200R2 and CD200R4. In contrast, an inhibitory receptor CD200R (also known as CD200R1) relies on other adaptors. CD200R3 is primarily expressed by basophils and mast cells, cells known to be involved in anti-parasitic immune responses and frequently also allergic reactions. The surface expression of CD200R3 decreases when basophils are activated by ligating their CD16 and CD32. Remarkably, most of this downregulation is observed within the first 30 minutes after activation. Unlike CD200R, CD200R3 does not bind CD200.
