Description: This three-color cocktail is designed to identify T cells from peripheral blood samples. It can be used alone, or in combination with other antibodies to further characterize T cell populations. The SK3 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by a majority of thymocytes, a subpopulation of mature T helper cells, and in low levels on monocytes. CD4 is a receptor for the human immunodeficiency virus (HIV). The SK7 monoclonal antibody reacts with human CD3 eplison, a 20 kDa subunit of the CD3/TCR complex. The CD3 complex, including the epsilon, gamma and delta subunits, is required for proper assembly, trafficking and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK1 monoclonal antibody reacts with the human CD8a molecule, an approximately 32-34 kDa cell surface receptor expressed either as a heterodimer with the CD8beta chain (CD8alpha beta) or as a homodimer (CD8alphaalpha). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. This cocktail contains the following antibodies: Anti-Human CD4 (SK3) FITC Anti-Human CD3 (SK7) PerCP-eFluor™ 710 Anti-Human CD8a (SK1) APC Applications Reported: eZfluor Anti-Human CD4 FITC, CD3 PerCP-eFluor™ 710, CD8a APC has been reported for use in flow cytometric analysis.

CD8 (Cluster of Differentiation 8) is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediate efficient cell-cell interactions within the immune system. The CD8 antigen acts as a co-receptor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. CD8 is found on a T cell subset of normal cytotoxic/suppressor cells which make up approximately 20-35 % of human peripheral blood lymphocytes. The CD8 antigen is also detected on natural killer (NK) cells, subpopulations of peripheral blood null cells, thymocytes and bone marrow cells. The CD8 co-receptor functions as either a homodimer composed of two alpha chains, or as a disulfide-linked heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. The majority of CD8+ T cells express CD8 as a alpha/beta heterodimer. In HIV, the HIV-2 envelope glycoprotein binds CD8 alpha chain (but not the beta chain).
