Mouse Monoclonal Antibody

The MAb PD7/26 recognizes CD45RB, which is an isoform of the Leukocyte Common Antigen (LCA) also known as CD45. CD45RB is a protein tyrosine phosphatase receptor that is present on most hematopoietic cells and absent on non-hematopoietic cells. Expression of CD45RB is more restricted than CD45 and can only be found on T cell subsets, B, NK, and myeloid cells. Regulatory T cells (Tregs) have been show to express low levels of CD45RB, which may correlate with increased migration to sites of infection. In contrast, high CD45RB expression results in IL-2 and IFN gamma production instead of IL-4 in vitro.

CD45RB is a receptor-type protein tyrosine phosphatase, CD45 glycoprotein. CD45 is crucial in lymphocyte development and antigen signaling, serving as an important regulator of Src-family kinases, promotes cell survival by modulating integrin-mediated signal transduction pathway and is also involved in DNA fragmentation during apoptosis. CD45 isoforms differ in their extracellular domains, but share identical transmembrane and cytoplasmic domains. CD45 isoforms differ in their ability to translocate into the glycosphingolipid-enriched membrane domains and their expression depends on cell type and physiological state of the cell. CD45RB is expressed in microglia and inflammatory cells. In addition to involvement in immunoreceptor signaling, CD45 is important in promoting cell survival by modulating integrin-mediated signal transduction pathway and is involved in DNA fragmentation during apoptosis. CD45 is expressed on cells of the human hematopoietic lineage with the exception of mature red cells.
