Mouse Monoclonal Antibody

Description: The monoclonal antibody OX26 reacts with rat CD71, which is also known as the transferrin receptor. This glycoprotein exists as a homodimer on the cell surface that facilitates the endocytosis of iron by binding to iron-saturated transferrin. CD71 is expressed by proliferating cells of hematopoietic lineages, but not by mature, resting cells or hematopoietic stem cells. Its increased expression, which can be induced by antigen and mitogen stimulation, has been shown to correlate with cell proliferation. Endothelial cells of brain capillaries, cortical neurons, oligodendrocytes, kidney tubules, hepatocytes, pancreatic cells, and muscle cells also express CD71. The OX26 antibody has been reported to exhibit agonist activity. Applications Reported: This OX26 antibody has been reported for use in flow cytometric analysis. Applications Tested: This OX26 antibody has been tested by flow cytometric analysis of rat bone marrow cells. This can be used at less than or equal to 0.25 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Excitation: 633-647 nm; Emission: 660 nm; Laser: Red Laser. Filtration: 0.2 μm post-manufacturing filtered.

Cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied transferrin receptor into specialized endosomes. Endosomal acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. Transferrin receptor is necessary for development of erythrocytes and the nervous system. Serum transferrin receptor (sTfR) is used as a means of detecting erythropoietin (EPO) misuse by athletes and as a diagnostic test for anemias resulting from a number of conditions including rheumatoid arthritis, pregnancy, irritable bowel syndrome and in HIV patients.
