Description: The monoclonal antibody J4-117 reacts with human CD72, a 45 kDa type II transmembrane glycoprotein that belongs to the C-type lectin superfamily. This glycoprotein is expressed on B cells at all stages of maturation except for plasma cells. CD72 is also expressed on dendritic cells, mast cells, and some macrophages. This receptor contains an immune-receptor tyrosine-base inhibition motif (ITIM) that associates with a tyrosine phosphatase, SHP-1. CD72 negatively regulates the major signal transduction pathways activated by the B cell receptor (BCR). Studies have demonstrated that the ligand for CD72 is CD100 (also known as Semaphorin 4D). CD72 can also react with CD5 to provide signaling between T and B cells. Finally, this receptor also prevents naive B cells from producing Ig and differentiating into B cells, and plays a role in controlling autoimmunity by enhancing apoptosis after Ag-specific BCR stimulation. Applications Reported: This J4-117 antibody has been reported for use in flow cytometric analysis. Applications Tested: This J4-117 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 μL (0.06 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. CD72 is a transmembrane glycoprotein expressed as a homodimer especially in B cells, but also in other antigen presenting cells such as dendritic cells and macrophages. Through one of its immunoreceptor tyrosine-based inhibitory motives (ITIMs), CD72 interacts with tyrosine phosphatase SHP-1, thereby suppressing B cell responsiveness. Binding of CD72 with its ligand CD100 (Sema4D) prevents BCR association and phosphorylation of CD72 and results in dissociation of SHP-1 from CD72, thus enables B cell activation.
