Description: This AD2 monoclonal antibody reacts with human CD73, a 5'-ectonucleotidase that converts 5'-adenosine monophosphate to adenosine. CD73 is expressed on the surface of endothelial cells, as well as B and T cells, including some CD4+Foxp3+ regulatory T cells. Adenosine production by these cells has been linked to the inhibition of CD4 T cell effector functions such as proliferation and cytokine secretion. Applications Reported: This AD2 antibody has been reported for use in flow cytometric analysis. Applications Tested: This AD2 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 μL (0.25 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. Light sensitivity: This tandem dye is sensitive photo-induced oxidation. Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-822-49) (100 μL cell sample + 100 μL IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. CD73 (Ecto-5-prime-nucleotidase, 5-prime-ribonucleotide phosphohydrolase) catalyzes the conversion at neutral pH of purine 5-prime mononucleotides to nucleosides, the preferred substrate being AMP. CD73 consists of a dimer of 2 identical 70 kDa subunits bound externally to the plasma membrane by a glycosyl phosphatidyl inositol linkage. CD73 is used as a marker of lymphocyte differentiation. Consequently, a deficiency of CD73 occurs in a variety of immunodeficiency diseases. Other forms of 5-prime nucleotidase exist in the cytoplasm and lysosomes and can be distinguished from CD73 by their substrate affinities, requirement for divalent magnesium ion, activation by ATP, and inhibition by inorganic phosphate. The CD73 gene has been localized to 6q14-q21 by immunofluorescence and a study of a panel of human x mouse hybrids that contained fragments of chromosome 6 as translocations. Defects in the CD73 gene can lead to the calcification of joints and arteries, and intestinal tuberculosis. Two transcript variants encoding different isoforms of CD73 have been found.
