General Description
- Formulated in 1966, Denhardt's solution is a combination of high-molecular weight polymers used to prevent non-specific binding of nucleic acid probes to membranes and filters
- Denhardt's solution contains 1% Ficoll (type 400), 1% polyvinylpyrrolidone, and 1% bovine serum albumin
Application
- Denhardt's solution is used in Northern and Southern blotting to prevent non-specific binding of detection reagents to membranes
- It can be used along with gelatin-chrome alum in in situ hybridization assays to maintain chromosome integrity
- It has been used to facilitate quantitative DNA hybridization on nylon membranes
