Grb2 protein contains one SH2 domain flanked on both sides by an SH3 domain. Ab-1 binds to the junction between the amino terminal SH3 domain and the SH2 domain of Grb2. Reactivity with Grb2 deletion mutants suggests that its epitope maps between aa 54-62. In response to EGF, Grb2 binds, through its SH3 domains, to the guanine nucleotide exchange factor, Sos. The Grb2-Sos complex then binds the EGF-receptor. These interactions play a critical role in the activation of the p21ras signalling pathway.
