The HIV-1 viral infectivity factor (Vif) is a crucial protein for the efficient replication of the virus, particularly in non-permissive cells such as lymphocytes and macrophages. Vif is a basic phosphoprotein with a molecular weight of approximately 23,000 Daltons. Its primary function is to counteract the host cellular antiviral protein APOBEC3G, which induces hypermutations in the viral DNA, thereby inhibiting viral replication. Vif achieves this by targeting APOBEC3G for ubiquitination and subsequent degradation, thus preventing it from incorporating into new virions. Structurally, Vif interacts with the cellular ubiquitin-proteasome machinery to facilitate the degradation of APOBEC3G. This interaction is essential for Vif's role in viral replication and immune evasion. Additionally, Vif has been shown to suppress the production of type I interferon by targeting the immune signaling molecule STING, further aiding in immune evasion.
