Biological Activity | Recombinant Human Wnt-3a induces beta-catenin responsive Topflash activation in HEK293T cells. The specific Wnt pathway inhibitor, endo-IWR 1, inhibits Topflash activity from a 2μg/mL dose of Recombinant Human Wnt-3a with a typical ED50 < 0.2μM. The less active diasteriomer exo-IWR 1 Wnt inhibitor shows minor inhibitory activity in this assay.Recombinant Human Wnt-3a induces alkaline phosphatase production in the MC3T3-E1 mouse pre-osteoblast cell line. The ED50 for this effect is typically 5-25ng/mL. The specific Wnt pathway inhibitor, endo-IWR 1, inhibits alkaline phosphatase production induced from a 50ng/mL dose of Recombinant Human Wnt-3a with an typical ED50 < 0.8μM. The less active diasteriomer exo-IWR 1 Wnt inhibitor shows a > 5μM ED50 with a Recombinant Human Wnt-3a dose of 50ng/ml. |
