Description
Protein IMPACT (UniProt Q9P2X3; also known as Imprinted and ancient gene protein homolog) is encoded by the IMPACT gene (Gene ID 55364) in human. IMPACT is a cytoplasmic protein preferentially expressed in neurons. IMPACT is highly abundant in the hypothalamus and in scattered neurons in other areas, such as the hippocampal interneurons. IMPACT and its yeast ortholog Yih1 contain an RWD domain (a.a. 14-116 of human and mouse IMPACT) that interacts with GCN1 and competes against GCN2 for GCN1 binding. IMPACT and Yih1 overexpression in mouse embryonic fibroblasts (MEFs) and yeasts inhibits GCN2 activation induction. IMPACT is a developmentally regulated protein that promotes neuritogenesis, while GCN2 strongly inhibits spontaneous neurite outgrowth. Increased abundance of IMPACT in differentiated neuronal cells enhances translation initiation by preventing GCN2 signaling.
