Mouse Monoclonal Antibody

The U31-644 monoclonal antibody specifically recognizes human Interferon regulatory factor 8 (IRF8, Interferon consensus sequence-binding protein, ICSBP or H-ICSBP). IRF8 is a ∽50kDa transcription factor that belongs to the interferon (IFN) regulatory factor family which consists of 9 members in humans and mice. IRF8 is variably expressed in hematopoietic cells including monocytes, macrophages, dendritic cells, and B cells. IRF8 has a conserved N-terminal DNA-binding domain (DBD) and a divergent C-terminal region that functions as the regulatory domain which specifies interactions between IRF8 and other IRF family members, transcription factors, or cofactors. IRF8 functions as a transcriptional activator or repressor depending on the formation of heterodimeric DNA binding complexes with these different partners. IRF8 binds to specific DNA motifs including IFN-stimulated response elements (ISRE) and regulates expression of genes including those that encode IFN-alpha (IFN-α), IFN-beta (IFN-β), and IFN-inducible MHC class I genes. It functions in innate and adaptive immunity by regulating the effector functions of monocytes/macrophages, B cells and dendritic cell subsets. IRF8 participates in the induction of Type I IFNs, TNF, IL-6, and IL-12 expression by leucocytes stimulated with ligands for Toll-like receptors and other pattern recognition receptors (PRRs). IRF8 is also involved in cell cycle regulations and apoptosis and may play a role in tumorigenesis.

R-phycoerythrin (PE) is an accessory photosynthetic pigment found in red algae. It exists in vitro as a 240-kDa protein with 23 phycoerythrobilin chromophores per molecule. This makes PE one of the brightest fluorochromes for flow cytometry applications, but its photobleaching properties make it unsuitable for fluorescence microscopy.
