Includes

Six PD-10 columns, filter paper, ethyl alcohol solution, Sephadex G-25, MSDS, methods of assessment manual, student data sheets and instructor manual

Requires

Coleus, beet, and spinach leaves, balance, lab glassware and supplies

Students learn how to separate pigments in plants.

Students pack Sephadex G-25 chromatographic column and calibrate it, then see a separation of anthocyanin, carotene, chlorophyll and xanthophyll from a mixture of these compounds. This separation is due to differences in molecular structures of these aromatic and cyclic compounds. The greater the number of rings in the pigments, the greater the affinity of the pigments for the gel granules.

  • Students separate pigments found in plants based on differences in molecular charge and structure
  • Students pack their own Sephadex G-25 chromatographic columns
  • Separation of anthocyanin, carotene, chlorophyll and exanthophyll from a mixture of plant pigments can be observed
  • Columns may be re-used
  • For grades 10 to 14
  • Class size: 24 students
