The Ragulator complex is comprised of three proteins, C11orf59, ROBLD3, and MAPKSP1, which targets mTORC1 to lysosomal surfaces. This normal endosomal trafficking is taken advantage of by certain viruses, including coronavirus, which has increased replication when C11orf59 is interfered with. C11orf59 is positively regulated by cellular cholesterol depletion and negatively regulated by cellular cholesterol loading. It is involved in LDL uptake and cholesterol homeostasis. C11orf59 and PPAR-alpha work together to control lipid and carbohydrate metabolism in diabetes. Nearly all effects of C11orf59 are associated with mTORC1 kinase growth promotion, and disregulation of the normal mTOR pathway.
