Cell surface glycoprotein MUC18 (UniProt P43121; also known as CD146, Cell surface glycoprotein P1H12, Melanoma-associated antigen A32, Melanoma-associated antigen MUC18, Melanoma cell adhesion molecule, S-endo 1 endothelial-associated antigen) is encoded by the MCAM (also known as MUC18) gene (Gene ID 4162) in human. MUC18/CD146 is an 113kDa transmembrane immunoglobulin superfamily glycoprotein mainly expressed on endothelial cells and smooth muscle cells where it functions as a cell-to-cell adhesion molecule, while little or no expression is found among epithelial cells and leukocytes (e.g. macrophages and neutrophils). Originally characterized as a protein overexpressed in malignant melanoma cells and adopted the name MCAM (melanoma cell adhesion molecule), MUC18/CD146 up-regulation is also reported in airway epithelial cells of patients with asthma and chronic obstructive pulmonary disease (COPD). Cellular overexpression and knockdown studies indicate that MUC18/CD146 plays a pro-inflammatory role in response to stimulation with various TLR agonists. MUC18/CD146 is produced with a signal peptide (a.a. 1-23) sequence, the removal of which yields the mature protein with a large extracellular region (a.a. 24-559), followed by a transmembrane domain (a.a. 560-583) and a cytoplasmic domain (a.a. 584-646). The extracellular region contains two V-type (a.a. 24-129 and 139-242) and three C2-type (a.a. 244-330, 335-424, 430-510) Ig-like domains.
