The transcription factor CCAAT/Enhancer Binding Protein beta (C/EBPβ, also known as NFIL6) is a highly conserved B-Zip family member. C/EBPβ regulates multiple genes involved in hepatic function, immune function and adipogenesis, and its functions are generally distinct from those of other C/EBP family members (alpha, gamma, delta, epsilon). C/EBPβ is present in cells as three alternate translation products: two transcription activating forms known as LAP1 and LAP2 (∽35 and 32kDa in rodents, ∽43kDa and 40kDa in humans) and a truncated inhibitory form known as LIP (∽20kDa) which lacks the N-terminal transcription activation domain. C/EBP β is phosphorylated at multiple sites which are determinants of some of its activities (e.g. activation of c-fos, adipogenesis). A recent report shows that it is acetylated at Lys215/216, and others report that it can be sumoylated. C/EBPβ is also acetylated at Lys39 in the transcription activation domain, and mutation of Lys39 impairs its ability to activate transcription of multiple genes, including several involved in adipogenesis.
