ERK3 (Extracellular signal-regulated kinase 3, MAPK6, p97 MAPK) belongs to the Ser/Thr protein kinase family and is a member of the mitogen-activated protein (MAP) kinase superfamily. Unlike the other members of the MAPK family, ERK3 is a mitogen-activated protein kinase homologue that has SEG motif instead of conserved TXY motif in the activation loop. Also, unlike ERK1 and ERK2, it is ubiquitously expressed and localized in the nucleus of proliferating cells. Highly unstable, ERK3 is constitutively degraded during ubiquitin-proteasome pathway of proliferating cells. ERK3 has been linked to phosphorylation MAP2 and as a possible regulator during cell differentiation.
