Nectin2 is modulator of T-cell signaling and can be either a costimulator of T-cell function, or a coinhibitor, depending on the receptor. Upon binding to CD226, Nectin2 stimulates T-cell proliferation and cytokine production, including that of IL2, IL5, IL10, IL13, and IFNG. Upon interaction with PVRIG, Nectin2 also inhibits T-cell proliferation. Nectin2 may play a role as a cell adhesion protein as one of the plasma membrane components of adherens junctions. Nectin2 also acts as a receptor for herpes simplex virus 1 (HHV-1) mutant Rid1, herpes simplex virus 1 (HHV-2) and pseudorabies virus (PRV). The gene for Nectin2 encodes a single-pass type I membrane glycoprotein with two Ig-like C2-type domains and an Ig-like V-type domain. Nectin2 also serves as an entry for certain mutant strains of herpes simplex virus and pseudorabies virus, and is involved in cell to cell spreading of these viruses. Variations in the Nectin2 gene have been associated with differences in the severity of multiple sclerosis. Alternate transcriptional splice variants of Nectin2 encoding different isoforms, have been characterized. Diseases associated with NECTIN2 include Herpes Simplex and Ovarian Cystic Teratoma.
