Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human PF-4 (CXCL4). Anti-Human PF-4 (CXCL4)-specific antibody was purified by affinity chromatography and then biotinylated. Sandwich ELISA: To detect Human PF-4 by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human PF-4 (500-P05) as a capture antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human PF-4. Western Blot: To detect hPF-4 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant hPF-4 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. 500-P05BT-1MG will be provided as 2 x 500 μg

Platelet factor-4 is a 70-amino acid protein that is released from the alpha-granules of activated platelets and binds with high affinity to heparin. Its major physiologic role appears to be neutralization of heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation. As a strong chemoattractant for neutrophils and fibroblasts, PF4 probably has a role in inflammation and wound repair (Eisman et al., 1990).
