Chromodomain helicase/ATPase DNA binding protein 1-like gene (CHD1L) is a ∼100.9kDa DNA helicase that is amplified in many solid tumors. The CHD1L gene, located at Chr 1q21 is frequently amplified in hepatocellular carcinoma. CHD1L plays a role in chromatin remodeling following DNA damage. It is targeted to DNA damage sites through interaction with poly(ADP)-ribose) and regulated chromatin during DNA repair. CHD1L contains a SNF2-N domain and a helicase superfamily domain, but unlike CHD1, it does not contain a chromodomain, but instead contains a c-terminal macrodomain, which is an (ADP)-ribose/ polymer of the ADP-ribose (PAR)-binding element. In addition to PAR binding, the macro domain of CHD1L is able to bind Nur77, a critical member of a p53-independent apoptotic pathway. This binding subsequently inhibits the nucleus-to-mitochondria translocation of Nur77 and disrupts apoptotic cell death. Overexpression of CHD1L protein in tumors is considered to be an independent biomarker of poor prognosis and short tumor-free survival time.
