The protein MHC class 1 polypeptide-related sequence A (MIC-A) and encoded by the human gene named MICA is an unusual MHC class one molecule with no known role in antigen presentation but instead acts as a stress-induced self antigen that is recognized by gamma delta T cells and is the ligand for KLRK1/NKG2D cellular receptor. Binding to KLRK1 leads to cell lysis. MICA is a cell membrane associated protein found in the cytoplasm. Additionally, it is expressed in many organ systems and found expressed on many cell types. Expression is induced by many stress responses, including infection, DNA damaging agents, E. coli contact, hCMV and ionizing radiation. MICA is also the antigen for many autoimmune diseases associated with the skin, one organ system where MICA is highly expressed. Psoriasis and Psoriatic arthritis are two such diseases and antibodies to MICA have also been associated with organ transplant difficulties and allograft rejection.
