Specifically detects phospho-AS160 (Thr642) in Human, Mouse samples, and it is validated for Western Blotting
AS160 is a widely expressed RAB-GTPase activating protein which plays a role in insulin action. AS160 is phosphorylated by AKT, AMPK, RSK, and SGK1. Upon phosphorylation, AS160 changes its location from microsomes to the cytosol upon insulin treatment. AS160 acts as key link between insulin signaling and GLUT4 trafficking of glucose. Phosphorylation of AS160 is essential for AS160 action and AS160 has multiple phosphorylation sites. Phosphorylation of AS160 on both AS160Thr642 and AS160Ser588 is important for insulin-stimulated glucose transport. Phosphorylation of these sites is predicted to inhibit AS160's activation of Rab-GTPase proteins and favor greater exocytosis of GLUT4 vesicles, leading ultimately to greater glucose transport. Research indicates that impairment of phosphorylation of AS160 at Thr642 is associated with various aspects of type 2 diabetes.
