Signal-peptide-peptidase-like 2A (SPPL2a), a member of aspartic protease family, is an ubiquitously expressed intramembrane protease that cleaves type II membrane signal peptides in the hydrophobic plane of the membrane. It has been implicated in the proteolysis of TNF-a, Fas Ligand, and Bri2. SPPL2a exhibits about 7-0% homology with SPPL2b, however, the later is expressed primarily in adrenal cortex and mammary gland. Over-expressed SPPL2a is observed in late endosomal compartments where it co-localizes with Rab7. Its lysosomal/late endosomal sorting is dependent on a single tyrosine-based motif (Tyr498) in its cytoplasmic tail.
