The 5H4 antibody monoclonal antibody specifically binds to mouse CD122. CD122 is a 70-85 kDa type I transmembrane glycoprotein of the hematopoietin receptor superfamily. CD122 is also known as IL-2 Receptor beta chain (IL-2R β ) or IL-15 Receptor beta chain (IL-15R β ) as it helps form signaling receptor complexes for Interleukin-2 (IL-2) or IL-15, respectively. CD122 can combine with either CD132 (γc) alone or CD132 plus CD25 (IL-2Rα) to form inte rmediate or high-affinity IL-2 Receptor complexes, respectively. The IL-2R β chain is constitutively expressed on NK cells and at lower levels on some thymocytes, T cells, B cells, monocytes, and macrophages. IL-2R β chain expression is up regulated by IL-2. The immunogen used to generate this hybridoma was rat myel oma cells transfected with a truncated IL-2R β cDNA.
The antibody was conjugated to BD Horizon™ BV650 which is part of the BD Horizon Brilliant™ Violet family of dyes. This dye is a tandem fluorochrome of BD Horizon BV421 with an Ex Max of 405-nm and an acceptor dye with an Em Max at 650-nm. BD Horizon BV650 can be excited by the violet laser and detected in a filter used to detect APC-like dyes (eg, 660/20-nm filter). Due to the excitation and emission characteristics of the acceptor dye, there will be spillover into the APC and Alexa Fluor™ 700 detectors. However, the spillover can be corrected through compensation as with any other dye combination.
