Perform the definative identification of yeast according to their ability to assimilate nitrogenous compounds with Thermo Scientific™ Remel™ Potassium Nitrate Assimilation Agar. This medium was developed by Wickerham to determine assimilation of nitrogen by yeast1. Wickerham found that inclusion of vitamins was necessary for certain yeast2. Pincus et al. modified the Wickerham formulation by adding higher concentrations of KNO3, yeast carbon base, pH indicator, and agar3.
Identify yeast according to their ability to assimilate nitrogenous compounds using the solid medium Potassium Nitrate Assimilation Agar. One criterion for the definitive identification of yeasts is the ability or inability to utilize an inorganic nitrogen source. Potassium nitrate is added to test the ability of the isolate to assimilate nitrogen. Bromthymol blue is the indicator used, which changes the medium from greenish yellow to blue-green or blue, indicating positive results.
Potassium Nitrate Assimilation Agar contains yeast carbon base, which provides amino acids, vitamins, trace elements and salts necessary to support growth of yeast. Noble agar is a pure, low ash agar, which is a solidifying agent in this medium.
