Differentiate Mycobacterium species based on urea hydrolysis with Thermo Scientific™ Remel™ Urea Broth For AFB. In 1979, Steadham developed the urease test to help in identification of mycobacteria1. This broth test helped to achieve more consistent results for urease activity. The determination of urea hydrolysis (release of ammonia) helps in the characterization of mycobacterial strains, in particular, to distinguish scotochromogens and nonphotochromogens.2-5
Urea Broth For AFB is a liquid medium used to detect Mycobacterium species based on urea hydrolysis. This is a highly buffered medium which helps in differentiation of mycobacterial strains after 7 days incubation.
Urea Broth For AFB contains peptone and dextrose that supply essential nutrients required for growth of mycobacteria. Urea acts as the substrate for hydrolysis. Polysorbate 80 is a surfactant which aids in the dispersion of aggregates. The low pH of 5.8 facilitates a clear-cut differentiation of positive and negative results. Phenol red (1%) serves as the pH indicator.
When urea is hydrolyzed ammonia is released and the resulting alkalinity is evidenced by the phenol red indicator. A dark pink or red color development is observed in the case of a positive test whereas broth remains yellow in the case of a negative test.
