ULP family members consist of a C-terminal core catalytic domain and N-terminal extensions determining substrate specificity. SENP8, a 24KDa protein, plays a major role in regulating NEDD8 pathway. It processes preNEDD8 to its mature form and deconjugates NEDD8 from substrates such as Cullins and p53. SENP8 may play a role in preventing accumulation of hyper-neddylated cullin and maintaining the level of mono-neddylated cullin that is thought to increase the efficiency of ubiquitination by SCF ubiquitin ligases.
