OMA1-06020 detects smoothelin from human, canine, feline, porcine, chicken and monkey tissues. This antibody detects both the visceral and vascular smoothelin isoforms. Does not cross react with rat samples. OMA1-06020 has been successfully used in Western blot, immunocytochemistry, and immunohistochemistry procedures. By Western blot, this antibody detects an ~59 kDa protein representing visceral smoothelin and an ~100 kDa protein representing vascular smoothelin from canine smooth muscle extracts. This antibody can be used to discriminate fully differentiated (contractile) smooth muscle cells from proliferative smooth muscle cells. Staining of formalin-fixed paraffin-embedded tissues requires boiling in 10 mM citrate buffer (pH 6.0) for 3x5 min, prior to immunostaining. OMA1-06020 antigen is cytoskeletal smoothelin extract of chicken gizzard. Store at 4ºC, or in small aliquots at -20ºC.

Smoothelin is a constituent of the smooth muscle cell (SMC) cytoskeleton. Antibodies directed to smoothelin are a useful tool to monitor SMC differentiation. Smoothelin is exclusively expressed in fully differentiated (contractile) SMCs. RNA and protein analyses reveal a broad species distribution of this protein. Cells with SMC-like characteristics, such as myofibroblasts, myoepithelial cells, and skeletal and cardiac muscle do not contain smoothelin. Confocal scanning laser microscopy of tissue sections, as well as transfection studies, show a filamentous organization of smoothelin that is different from that of desmin and vimentin. Smoothelin colocalizes with actin stress fibers. Two tissue specific isoforms have been identified: a 59 kDa isoform specific for visceral SMC; and a 100 kDa specific for vascular SMC. Human smoothelin is encoded by a single copy gene which is located on chromosome 22.
