The TaqMan Cells-to-CT HepatoExpress Kit offers a complete cell lysis and RT-qPCR workflow solution designed for high-throughput gene expression analysis directly from cultured primary hepatocytes without the need for RNA purification. This innovative kit includes HepatoExpress Lysis Solution, HepatoExpress LysisGuard reagent, ezDNase enzyme, SuperScript IV VILO Master Mix, and the TaqMan Fast Advanced Master Mix. The newly developed HepatoExpress Lysis Solution and LysisGuard reagents enable superior lysis performance and sample stability, while the best-in-class RT-qPCR reagents deliver high sensitivity for a seamless end-to-end workflow in approximately 75 minutes.

Optimized chemistry without sacrificing sensitivity

Gene expression analysis from primary hepatocytes is now simplified with our TaqMan Cells-to-CT HepatoExpress Kit. With a strong lysis that penetrates extracellular matrix overlays without degrading RNA, this product enables a rapid workflow without compromising sensitivity. The inclusion of ezDNase enables integrated removal of genomic DNA during cell lysis. After a 10-minute incubation at room temperature, the lysate is ready for reverse transcription using SuperScript IV VILO Master Mix. Once the RT reaction is complete, qPCR analysis is conducted using the TaqMan Fast Advanced qPCR Master Mix and a TaqMan Gene Expression Assay.

Streamlined workflow with proven performance

RNA prepared using the TaqMan Cells-to-CT HepatoExpress technology substantially expedites and streamlines gene expression analysis of primary hepatocytes. Cell lysates exhibit sensitivity and specificity equivalent to RNA extracted using traditional spin column isolation methods.

The lysis procedure simultaneously prepares cell lysates for RT-qPCR and removes genomic DNA in only ten minutes. The lysis step is easily automated with robotic platforms for high-throughput processing of 96-well plates, as it occurs entirely at room temperature. The high-efficiency SuperScript IV VILO RT Master Mix linearly transcribes RNA across a broad input range, even with suboptimal template purity. Additionally, the TaqMan Cells-to-CT HepatoExpress Kit enables multiplexed detection of targets and is compatible with over 700,000 TaqMan Gene Expression Assays.
