Description
TBE (Tris/Borate/EDTA) Buffer is commonly used in nucleic acid electrophoresis. TBE Buffer is used for polyacrylamide and agarose gel electrophoresis. It is ideally suited for DNA and RNA separation on longer run, higher voltage or amperage gels. TBE is used with non-denaturing or denaturing (7 M urea) gels. It is also routinely used for DNA automated sequencing gel.5 TBE can also be used for agarose gels but is not recommended for preparative gels for recovery of nucleic acids. Tris-Borate-EDTA buffer has been used for pulsed-field gel electrophoresis (PFGE).

Concentrations of 5X Tris Borate EDTA (TBE) buffer or greater may precipitate due to the nature of the product. This precipitation is a result of nucleation of salt crystals by dust particles or other insoluble materials. In some cases, we recommend heating the buffer to ~ 50°C to re-dissolve the TBE into solution. Alternately, to avoid precipitation of high concentration TBE buffers, after the solution is prepared, you can filter it through a 0.2 µm cellulose acetate or cellulose nitrate filter. These treatments have no deleterious effect on the buffering properties of TBE.
