Novel thermostable DNA polymerase that uses dideoxynucleotide triphosphates (ddNTPs) as readily as deoxynucleotide (dNTP) substrates

  • Uniform and easy-to-read band patterns
  • Better incorporation of ddNTPs helps avoid secondary structures that can cause unreadable sequences
  • Read-through of difficult sequences (e.g., GC-rich), due to the ability of thermostable enzymes to function at elevated temperature used for sequencing
  • ddNTPs are chain-elongating inhibitors of DNA polymerase, which are used in the Sanger method for DNA sequencing
  • Each nucleotide base of this particular type has a probability of being bonded not to a deoxynucleotide but rather a dideoxynucleotide, which ends chain elongation
  • An excellent choice for cycle sequencing where template quantity limitations preclude the use of T7 Sequenase DNA polymerase
