TSKgel™ Phenyl-5PW RP columns are filled with 10 micron porous polymethacrylate particles, the surface of which is covered with a high density of phenyl groups that causes proteins to be retained in a low salt buffer mobile phase and requiring a solvent gradient for their elution, as is the protocol in reversed phase chromatography.

In contrast, TSKgel™ Phenyl-5PW has such a low density of phenyl groups on the surface that proteins can only be retained at high salt concentration and eluted by a decreasing salt gradient, which, of course, is the protocol used in hydrophobic interaction chromatography. TSKgel™ Phenyl-5PW RP columns can withstand rigorous cleaning with either acid or base. The 1000 Å pore size accommodates globular protein samples up to 1,000,000 Da.
