The TTLL10 gene encodes a protein that functions as a polyglycylase, playing an important role in the modification of nucleosome assembly protein 1 (NAP1) both in vivo and in vitro. This enzymatic activity is dependent on the presence of ATP and is abolished in mutant forms where the glutamate in the active site is replaced by valine. TTLL10 specifically incorporates glycine into recombinant NAP1 at Glu residues 359 and 360. This modification process likely has implications for protein interactions and cellular processes involving NAP1. Additionally, TTLL10 can perform tubulin glycylation when co-expressed with TTLL8, suggesting a complex interplay between these enzymes in cellular microtubule dynamics.
