Reverse transcription polymerase chain reaction (RT-PCR) is a laboratory technique that combines the reverse transcription of RNA into DNA and the subsequent amplification of the DNA using PCR.
In RT-PCR, a reverse transcriptase enzyme converts an RNA template to its complementary DNA (cDNA). The cDNA then becomes the template for PCR amplification.
A typical RT-PCR kit includes the main components needed for the assay:
- Buffer
- Primers – a combination of deoxynucleotide triphosphates (dNTPs) of the four bases of adenine (dATP), cytosine (dCTP), guanine (dGTP), and thymine (dTTP)
- Reverse transcriptase reagent
The combination of RT-PCR and qPCR techniques (sometimes labelled RT-qPCR) is often used in research and clinical labs to analyze gene expression and quantify viral RNA. Since this method uses fluorescence to track the amplification reaction, these kits may also include:
- Fluorogenic primers or labeled fluorogenic probes
- Fluorescent dye
Reagents for cDNA PCR can be purchased separately or as cDNA synthesis kits.
RT-PCR is commonly used for RNA detection, molecular cloning, and gene sequencing. RT-qPCR is used for gene expression analysis, RNAi or microarray validation, pathogen detection, genetic testing, and other disease research.

Meridian’s universal Inhibitor-Tolerant qPCR is a 2x formulation ready-to-use quantitative PCR supermix, containing all components except primers, probes and templates for direct qPCR. It is designed for developing qPCR multiplex assays that require minimal sample processing and fast turn-around times (TAT). Because the mix is highly resistant to various qPCR inhibitors, it can be used with crude lysates or inhibitor-rich samples to directly detect pathogens, including viruses and bacteria, in various biological specimens and to analyze gene expression in cells.
Meridian Life Science Low DNA qPCR Mix, 5ML
Low DNA qPCR master mix contains a chemical hot-start Taq polymerase and highly optimized reaction buffer with dNTPs, MgCl2, stabilizers and PCR enhancers. The Taq polymerase is purified twice, before and after the addition of the chemical hot-start, this helps to remove trace amounts of DNA (known as bioburden) that come from the manufacturing of the DNA polymerase. In addition there is no bioburden that comes with antibody hot-start, making it ideal for low-copy number targets and also microbial DNA and fungal DNA detection assays. Low DNA qPCR master mix can also be used for fast, precise and highly reproducible genotyping of sequence variants, including loci, provides higher stringency and more specific binding of the allele-specific probes, with wider and clearer separation of allele cluster.

Meridian Life Science RT-qPCR Extraction Control, 1ML
Molecular IVD companies continue to experience tighter and tighter regulations in regard to incorporating appropriate positive and negative controls into their assays. Meridian’s RT-qPCR Extraction Control Orange uses artificial micelles that contain quantitative PCR positive control RNA sequence (with no known homology to any organism, so that it does not interfere with the detection of the sample target RNA). The cells are added to the RT-qPCR Extraction Control is spiked in the with lysis buffer into the sample prior to RNA extraction for downstream amplification by RT-qPCR. The detection of RT-qPCR Extraction Control acts as a cDNA synthesis and PCR positive control and confirms the success of the extraction and amplification steps, avoiding the misinterpretation of a false negative result.

The Luna Probe One-Step RT-qPCR Mix with UDG (No ROX) is supplied at a 4X concentration and enables higher amounts of sample input which is relevant for applications where RNA present in low abundance is of interest such as pathogen detection. The mix consolidates the components for one-step RT-qPCR into a single tube including Luna WarmStart RT Hot Start Taq DNA Polymerase dNTPs and Murine RNase Inhibitor in an optimized buffer. Combining Hot Start Taq DNA Polymerase with a novel WarmStart-activated reverse transcriptase allows for dual control of enzyme activity via reversible aptamer-based inhibition. This temperature-dependent activation helps to prevent undesirable non-specific priming and extension prior to thermocycling providing added security for setting up reactions at room temperature. This formulation contains no reference dye and is compatible with any instrument that does not require ROX (if ROX normalization is needed ROX can be added.)
