The PathHunter mFFAR1 cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. free fatty acids) based activation of mFFAR1 GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged mFFAR1 and EA-tagged β-Arrestin-2. Activation of mFFAR1 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. This cell line enables assessment of ligand (e.g. relaxin-1) based activation of RXFP2 GPCR activity via detection of increasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type RXFP2, a naturally Gs coupled GPCR. Upon activation of RXFP2, the increase in levels of intracellular cAMP leads to an increased signal from EFC when the cAMP detection kit is applied. This cell line enables assessment of chaperone-based stabilization of CFTR-ΔF508 via detection of rescued membrane trafficking. The included cell line overexpresses ProLink-tagged mutant CFTR-ΔF508, which is misfolded and retained in the ER. The cell line also expresses membrane-localized EA. When a pharmacochaperone stabilizes the mutant CFTR, it is trafficked to the membrane, producing EFC signal. This stable modified cell line is provided as two vials of cryopreserved cells. RM-1 is a cell line with fibroblast-like morphology that was isolated from the prostate of a 17-day-old mouse. 17-day-old urogenital sinus cells of C57BL/6 embryos were infected with Zipras/myc9 retrovirus and grafted under the renal capsule of isogenic adult male mice. It was deposited by Baylor College of Medicine.
