Annexin V-fITC kit allows fluorescent detection of annexin V bound to apoptotic cells and quantitative determination by flow cytometry. The AnnexinV-fITC kit uses annexin V conjugated with fluorescein isothiocyante (fITC) to label phosphatidylserine sites on the membrane surface. The kit includes propidium iodide (PI) to label the cellular DNA in necrotic cells where the cell membrane has been totally compromised. This combination allows the differentiation among early apoptotic cells (annexin V positive PI negative) necrotic cells (annexin V positive PI positive) and viable cells (annexin V negative PI negative).

It inhibits ubiquitin-specific-processing protease 14 USP14 and ubiquitin c-terminal hydrolase isozyme L5 UCHL5 UCH37, two 19S regulatory particle proteasome-associated DUBs. It exhibits minimal inhibition of UCHL-1, UCHL-3, USP2, USP7, USP and BAP1. WP1130 is a second-generation tyrphostin derivative initially identified as a JAK2 signaling inhibitor through JAK2 ubiquitination that reduces STAT activation stimulated by cytokines such as IL-6 and IL-3 through the rapid down-regulation of upstream JAK kinases. WP1130 has also been identified as a partially selective cell permeable deubiquitinating enzyme/deubiquitinase DUB inhibitor that directly inhibits DUB activity of USP9x, USP5, USP14, USP24, UCH37, UCHL1 and UCHL5, leading to rapid accumulation of polyubiquitinated K48/K63-linked proteins into juxtanuclear aggresomes and tumor cell apoptosis, accounting for its anti-cancer properties. Through this mechanism, WP1130 has been shown to downregulate the antiapoptotic proteins Bcr/Abl and Jak2 and to upregulate the proapoptotic proteins MCL-1 and p53.

The Apoptosis and Necrosis Quantitation Kit Plus is a simple, convenient assay for quantifying apoptotic (green; Abs/Em: 490/515 nm) and necrotic (red; Abs/Em with DNA: 528/617 nm) cells within the same cell population by flow cytometry or fluorescence microscopy. Apoptosis is accompanied by phosphatidylserine (PS) translocation from the inner to the outer leaflet of the plasma membrane. Exposed PS is recognized by Annexin V, a calcium-dependant high-affinity phospholipid-binding protein. CF488A conjugated Annexin V stains apoptotic cells green. CF488A is spectrally similar to fluorescein (FITC), but is brighter and more photostable. Necrosis results from severe cellular injury causing loss of plasma membrane integrity. Ethidium homodimer III (EthD-III), a membrane impermeant high-affinity DNA binding dye stains cells with compromised membranes red. It is superior option to propidium iodide (PI) or ethidium homodimer I due to higher DNA affinity and fluorescence quantum yield.
