This gene encodes paired box gene 6, one of many human homologues of the Drosophila melanogaster gene prd. In addition to the hallmark feature of this gene family, a conserved paired box domain, the encoded protein also contains a homeo box domain. Both domains are known to bind DNA, and function as regulators of gene transcription. This gene is expressed in the developing nervous system, and in developing eyes. Mutations in this gene are known to cause aniridia as well as Peter's anomaly, both ocular diseases

This protein has been shown to play roles in platelet aggregation, angiogenesis, and tumorigenesis.

Epac (exchange protein directly activated by cAMP; also designated cAMP-GEFI) and Epac2 (also designated cAMP-GEFII) compose a novel family of cyclic AMP activated guanine-nucleotide exchange factors (GEFs). Both Epac and Epac2 contain cAMP-binding motifs and a motif common to Ras superfamily GEFs. Epac and Epac2 both function to activate Rap1, a Ras-like GTPase that is known to suppress the oncogenic transformation of cells by Ras. Epac is ubiquitously expressed, whereas Epac2 is primarily expressed in the brain and adrenal glands.

Mammalian homologs of the Drosophila Mad gene include Smad1 (also designated Madr1 or JV4-1), Smad2 (also designated Madr2 or JV18-1), Smad3, Smad4 (also designated DPC4), Smad5, Smad6, Smad7 and Smad8 (also designated Smad9). Smad1 and Smad5 are effectors of BMP2 and BMP4 function while Smad2 and Smad3 are involved in TGFb and activin-mediated growth modulation. Smad4 is required for all of the above activities and necessitates FAST-1 for DNA binding. Smad6 and Smad7 regulate the response to activin/ TGF-beta signaling by interfering with TGF-beta-mediated phosphorylation of other Smad family members.

The protein encoded by this gene is a member of a family of proteins which inhibit apoptosis through binding to tumor necrosis factor receptor-associated factors TRAF1 and TRAF2. Similar to API1, BIRC4 inhibits apoptosis induced by menadione, a potent inducer of free radicals, and ICE. BIRC4 also inhibits at least two members of the caspase family of cell-death proteases, caspase-3 and caspase-7.

Interferon-gamma (IFN-γ), also known as Type II interferon or immune interferon, is a cytokine produced primarily by T-lymphocytes and natural killer cells. The protein shares no significant homology with IFN-β or the various IFN-α family proteins. IFN-γ was originally characterized based on its antiviral activities. The protein also exerts anti-proliferative, immuno-regulatory and pro-inflammatory activities and is thus important in host defense mechanisms. IFN-γ exerts its biological activities by binding to specific cell surface receptors with high-affinity binding sites.

Cas9 is the nuclease guided by the crRNA and tracrRNA (or trans-activating crRNA) to cleave specific DNA sequences. A guide RNA (gRNA) can be designed to include a hairpin that mimics the tracrRNA-crRNA complex. Binding specificity is based on the gRNA and a three nucleotide NGG sequence called the protospacer adjacent motif (PAM) sequence. With its ease in designing guide sequences to target specific genomic loci, the CRISPR/Cas system is a much simpler, faster, and robust alternative to TALEN and Zinc finger nuclease platforms. For site-specific genome editing, the CRISPR/Cas9 system requires the Cas 9 nuclease and the gRNA. This adenovirus expresses a codon-optimized Cas9 nuclease, along with a mCherry reporter. It can be used along with guided RNA for genome-editing.This adenovirus expresses a FLAG tagged Cas9 with a mCherry reporter.
