Understanding lysosome-related physiology requires lysosome-specific probes that can track the behavior of lysosomes within cells. As a consequence of being designed simply to target acidic environments, most lysosomal probes have poor specificity towards the lysosome and stain many non-lysosomal acidic compartments. Moreover, existing lysosome probes’ fluorescent intensity is highly reliant on the lysosomes’ pH; their emission intensity is unstable and varies significantly due to differences in acidity among lysosomes, even within a cell. Biotium MB succinimidyl ester
The amine reactive succinimidyl ester of MB (methylene blue) can be conjugated to peptides, proteins, drugs, polymeric materials and biomolecules with primary amine groups. The conjugate will have a blue color and be able to complex with nucleic acids. Methylene blue is a well known redox dye which can be reversibly reduced to its colorless leuco form. Upon oxidation, the blue color of the dye is recovered.

IRDye infrared dyes from LI-COR Biosciences are available for researchers developing applications using near-infrared (NIR) fluorescence. These unique dyes are used in a variety of NIR imaging applications including Western blotting, plate-based assays, protein arrays, tissue section imaging, and molecular activity measurements. Standard NHS ester chemistry is used to produce custom probes labeled with LI-COR IRDye infrared dyes. The NHS ester reactive group provides the functionality for labeling primary and secondary amines, such as lysine residues in proteins.

MitoView™ dyes are membrane-permeant fluorogenic stains that rapidly accumulate in mitochondria of live cells. They can be imaged without washing, and are available with blue, green, far-red, and near-infrared fluorescence.MitoView™ Green is a green fluorescent mitochondrial dye (Ex/Em 490/520 nm) similar to MitoTracker Green FM. The dye is non-fluorescent until it paritions into the mitochondrial membrane. The staining relies on mitochondrial mass, not on mitochondrial membrane potential (MMP) and can therefore be used in both live cells and fixed cells. SDA azide contains both diazirine photoaffinity moiety and azide functional group. It is an excellent building block to introduce photoaffinity function into an alkyne-modified biomolecule via the well-known click chemistry. Diazirines are known for their ability to undergo photochemical reactions when exposed to ultraviolet (UV) light, specifically by forming highly reactive carbene intermediates that react with nearby molecules, forming covalent bonds. This property makes diazirines useful for studying protein-protein, protein-nucleic acid interactions, ligand-receptor binding, and other biomolecular interactions. It's important to note that diazirines are highly reactive and can be challenging to handle due to their instability. They require careful storage and handling, typically in a controlled environment.
