Recognizes a protein of 55 kDa, identified as CD25 (Workshop IV; Code A27). CD25 is expressed on activated T- and B-cells and activated monocytes/macrophages. With respect to lymphomas, CD25 is present on malignant cells of Hodgkin's disease, HTLV-1 adult T-cell leukemia, cutaneous T-cell lymphoma, and hair cell leukemia. Increased levels of soluble CD25 are observed in the leukemias/lymphomas and inflammatory/ autoimmune diseases. CD25 molecule alone appears to function as a low affinity receptor and associates with CD122 (IL-2R beta chain, p75) and CD132 (common gamma chain) to form the high affinity IL-2 receptor complex. CD25 antibodies detect three epitope regions, A, B and C. This MAb reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Biotium MUC2(CCP58), CF568 conjugate, 0.1mg/mL
Recognizes a single glycoprotein of 520 kDa, identified as mucin 2 (MUC2). This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC3. Its epitope has been defined as GTQTP (GlyThrGlnThrPro). Mucins are high molecular weight glycoproteins, which constitute the major component of the mucus layer that protects the gastric epithelium. MUC2 is specifically expressed in goblet cells of the small intestine & colon; in about 65% of colonic carcinomas and about 40% of gastric carcinomas. This MAb reacts with a protein of 22 kDa, identified as β sub-unit of HCG. It does not cross react with the α sub-unit. HCG is a glycoprotein, which is secreted in large quantities by normal trophoblasts. It is present only in trace amounts in non-pregnant urine and sera but rises sharply during pregnancy. HCG is composed of two non-identical, non-covalently linked polypeptide chains designated as the alpha and beta subunits. The alpha subunit is identical to that of thyroid stimulating hormone (TSH), follicle stimulating hormone (FSH), and luteinizing hormone (LH). hCG MAb detects cells and tumors of trophoblastic origin such as choriocarcinoma. Ten to forty percent of the patients with acquired immunodeficiency syndrome (AIDS) develop cytomegalovirus (CMV) infections. In some patients with AIDS, CMV is detected in the bronchoalveolar lavage fluid (BALF), urine, and other specimens, even when there are no symptoms of CMV disease. An indicator of active CMV infection is needed to facilitate the diagnosis of CMV disease in patients with AIDS or HIV infection. CD19 is a transmembrane glycoprotein that contains two extracellular immunoglobulin-like domains. CD19 is present in both benign and malignant B-cells and is considered to be the most reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells, and in scattered cells of the inter-follicular areas. Anti-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However, in contrast to CD20, expression of CD19 is continuous throughout B-cell development and through terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cell neoplasms commonly at a lower intensity than normal B-cell counterparts. Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8 T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs). CD117, also known as c-kit, is a member if the immunoglobin superfamily. It is a receptor tyrosine kinase that is expressed on hematopoietic progenitor cells. Protein phosphorylation is a fundamental event in the regulation of a large number of intracellular processes. Phosphorylation of specific tyrosine residues is the result of activation or stimulation of their respective protein tyrosine kinases. The phosphorylated proteins can be auto-phosphorylated kinases or certain cellular protein substrates. Tyrosine-phosphorylated proteins are involved in signal transduction and in the regulation of cell proliferation. Antibody to phosphotyrosine provides an excellent tool for the detection, characterization, and purification of phosphotyrosine containing proteins. Expression of NKX2.2 has been found in neuroendocrine tumors of the gut, making it a potential marker for the study of gastrointestinal neuroendocrine tumors. More recently, NKX2.2 protein was identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma, and was shown to be differentially upregulated in Ewing sarcoma on the basis of array-based gene expression analysis.
