Cayman’s L-Lactate Assay provides both a fluorescence and an absorbance-based method for detecting L-lactate in biological samples such as serum, plasma, blood, urine, and saliva. It can also be utilized to determine intracellular and extracellular lactate concentrations in cell culture samples. In the assay, LDH catalyzes the oxidation of lactate to pyruvate, along with the concomitant reduction of NAD+ to NADH. NADH reacts with the substrate to yield a final product that can be measured fluorometrically with an excitation wavelength of 530-540 nm and an emission wavelength of 585-595 nm, or colorimetrically at 535 nm.

Cayman’s Superoxide Dismutase Assay Kit provides a convenient method of measuring SOD activity in plasma, serum, erythrocyte lysates, tissue homogenates, and cell lysates. The assay relies on xanthine oxidase to produce superoxide radicals, which then reduce a tetrazolium salt to a formazan dye, detectable at 450 nm. SOD in the sample inhibits this reduction by catalyzing the dismutation of superoxide radicals. Thus, the degree of inhibition is directly proportional to SOD activity. The SOD Assay Kit measures all three types of SOD (Cu/Zn-, Mn-, and Fe-SOD). Mitochondrial Mn-SOD can be assayed separately following the procedure outlined in the kit booklet.

Cayman’s cGAS Inhibitor Screening Assay Kit provides a robust and EAsy-to-use platform for identifying novel inhibitors of human cGAS, which is a cytosolic dsDNA sensor that detects microbial pathogens. The assay directly measures 2’3’-cGAMP produced by cGAS in the presence of DNA, ATP, and GTP. The cyclic dinucleotide product of that reaction is quantified via ELISA using a 2’3’-cGAMP-specific antiserum. The cGAS inhibitor CU-76 is included as a positive control. The ELISA portion of Cayman’s cGAS Inhibitor Screening Assay Kit is a competitive assay with a range of 4.57-10,000 pM, a midpoint of approximately 300 pM (50% B/B0), and a sensitivity (80% B/B0) of approximately 50 pM.

Cell Biolabs’ NAD+/NADH Assay Kit (Colorimetric) is a simple assay that measures NAD+ and NADH present in biological samples such as cell lysates or tissue extracts in a 96-well microtiter plate format. The kit is specific for NAD+, NADH, and their ratio. The kit will not detect NADP+ or NADPH. The total NAD+/NADH concentrations of unknown samples are determined by comparison with a known NAD+ standard. Determination of both NAD+ and NADH requires two separate samples for quantification. NAD+ and NADH do not need to be purified from samples, but rather can be extracted individually with a simple acid or base treatment prior to performing the assay. The kit has a detection sensitivity limit of approximately 4 nM NAD+. Each kit provides sufficient reagents to perform up to 100 assays, including blanks, NAD+ standards and unknown samples.
Cell Biolabs Inc DAG (Diacylglyercol) Assay Kit, 100 assays
Diacylglycerols (DAG) are key intermediates in the biosynthesis of many cellular lipids and play a fundamental role in biochemical signaling. Cell Biolabs’ DAG Assay Kit simple 96-well fluorometric assay that measures diacylglycerol content in samples by a coupled enzymatic reaction system. First, kinase is used to phosphorylate DAG samples, yielding phosphatidic acid. Next, a lipase is used to hydrolyze phosphatidic acid to glycerol-3-phosphate. Finally, the glycerol-3-phosphate product is oxidized by glycerol-3-phosphate oxidase (GPO), producing hydrogen peroxide which reacts with the kit’s fluorometric probe. The kit contains a DAG Standard and has a detection sensitivity limit of 15 µM. Each kit provides sufficient reagents to perform up to 100 assays, including blanks, standards and unknown samples. Note each sample replicate requires 2 assays, one of which is a negative control to be subtracted as background.
