Elevated glucose uptake is a marker of tumors, and glucose transporters are important targets in cancer treatment. One common method for evaluating the glucose uptake ability of cells uses radioisotope-labeled glucose. Although this method has been used for many years, it requires special handling facilities and disposal of radioactive materials, at the disadvantage of inconvenience and high cost. 2-NBDG has been used widely to detect cellular glucose uptake, but the sensitivity is poor because of the low fluorescence intensity.Glucose Uptake Probe-Green has resolved those limitations. This probe emits strong green fluorescence (Ex=507 nm, Em=518 nm), allowing for easy detection of glucose uptake over 200 times more sensitive than 2-NBDG by fluorescent microscope, microplate reader, and flow cytometry.The WI Solution in this kit enhances cellular retention of the probe to give more reliable data. This gene encodes pyruvate carboxylase, which requires biotin and ATP to catalyse the carboxylation of pyruvate to oxaloacetate. The active enzyme is a homotetramer arranged in a tetrahedron which is located exclusively in the mitochondrial matrix. Pyruvate carboxylase is involved in gluconeogenesis, lipogenesis, insulin secretion and synthesis of the neurotransmitter glutamate. Mutations in this gene have been associated with pyruvate carboxylase deficiency. Alternatively spliced transcript variants with different 5 UTRs, but encoding the same protein, have been found for this gene.
