The SUMO-QAPTURE-T kit is an efficient tool for the selective isolation of SUMOylated proteins. The kit facilitates the affinity purification of SUMOylated proteins from cell extracts and tissue lysates using a high-binding SIM-containing affinity matrix. Captured proteins are eluted under denaturing conditions followed by analysis by Western blotting, using the SUMO antibodies provided or antibodies to specific proteins of interest, or potential substrate identification by proteomic methods. SUMO-conjugate-containing samples are prepared in native form in the presence of protease inhibitors to prevent loss through the action of deSUMOylating enzymes. Optimization of binding permits complete isolation of the full range of SUMO-protein conjugates from a specific lysate.

This MAb is a very special because it reacts ONLY with the intact-HCG and not with either free alpha- or free beta-chain of HCG. HCG is a glycoprotein and is composed of two non-identical, non-covalently linked polypeptide chains designated as the alpha and beta subunits. The alpha subunit is identical to that of thyroid stimulating hormone (TSH), follicle stimulating hormone (FSH), and luteinizing hormone (LH). HCG is secreted in large quantities by normal trophoblasts. It is present only in trace amounts in non-pregnant urine and sera but rises sharply during pregnancy. HCG MAb detects cells and tumors of trophoblastic origin such as choriocarcinoma. Large cell carcinoma and adenocarcinoma of the lung demonstrate anti-hCG positivity in 90% and 60% of cases respectively. 20% of lung squamous cell carcinomas are positive. Recombinant SUMOstar Protease 1 specifically recognizes and cleaves the SUMOstar tag from recombinant fusion proteins made with SUMOstar expression systems (Cat.#s: 1100A, 1106, 1100K, 1101, 2100, 2101, 2160, 2161, 3100, 3101, 7110, 7111, 7120, 7121). Unlike thrombin, EK, or TEV proteases, which recognize short, linear sequences, SUMOstar Protease 1 recognizes the tertiary structure of SUMOstar. As a result, SUMOstar Protease 1 will not cleave within the protein of interest. SUMOstar Protease 1 will not cleave SUMO-tags from proteins expressed with the SUMOpro or SUMOpro3 systems. To cleave the SUMOpro tag (orginial yeast SUMO tag), please use SUMO Protease 1 and to cleave SUMOpro3 tag please use SUMO Protease 2. This product contains 5000 units of SUMOstar Protease, where 1 U will cleave >90 µg of SUMOstar-GFP in 1 hour at 37C
