This retrovirus titer boost is a novel recipe of small molecules designed for increasing / boosting retrovirus packaging efficiency. Retrovirus 10X Titer-Up stably regulates the viral RNA packaging on the transcriptional level, which can enhance production of retroviral particles for up to 10 fold.

Instead of using lengthy spins in an ultracentrifuge, Retro-Concentin enables concentration of retroviruses directly from culture medium with a simple one (or an optional two) low-speed centrifugation steps. In addition, Retro-Concentin stabilizes the retroviruses for frozen storage, providing an additional advantage over other concentration methods. The rate at which retroviral vectors bind to and infect cells is mostly controlled by diffusion. During retrovirus infection, only a small fraction of the retroviral vectors can transduce target cells. Virion adsorption is the limiting step of this process. The use of polycations, such as Polybrene, is standard in many retroviral infection protocols owing to the observations of improved infection efficiency. Its mechanism of action is thought to involve neutralization of electrostatic repulsion between virion and cell membranes, enhancing attachment. ViraDuctin™ Retrovirus Transduction Kit is a proprietary formulation for the transduction of retrovirus. The KILR Retroparticles for Suspension Cells (G418) are MMLV particles designed to transduce suspension cells. This allows production of ProLabel-tagged, G418-resistant target cells of interest for use in cytotoxicity assays involving detection of cell lysis. This product uses EFC technology. The retroparticles mix can be used to transduce target cells with a ProLabel-tagged housekeeping gene. Target cell death in a cytotoxicity assay will release ProLabel to the media, where EA contained in the detection reagent will produce EFC signal.
