Although the specificity of Maackia amurensis lectin II (MAL II) is not well defined, it appears to bind only carbohydrate structures that contain sialic acid. Unlike Sambucus nigra lectin (SNA), which prefers structures with (α-2,6) linked sialic acid, MAL II binds sialic acid in an (α-2,3) linkage. Tissue staining patterns are also different among MAL I, SNA, and MAL II.

Sambucus nigra agglutinin (SNA) is a lectin isolated from elderberry bark that binds preferentially to sialic acid attached to terminal galactose in α-2,6 and to a lesser degree, α-2,3 linkage. Binding is also inhibited to some extent by lactose or galactose. This lectin does not appear to bind sialic acid linked to N-acetylgalactosamine. SNA has been reported to inhibit cell-free protein synthesis. This Cy5-labeled SNA features a ratio of fluorochrome to lectin that provides optimal staining (excitation 649 nm, emission 670 nm, color far red) and is supplied essentially free of unconjugated fluorochrome.

VVL recognizes preferentially α- or β-linked terminal N-acetylgalactosamine, especially a single α-N-acetylgalactosamine residue linked to serine or threonine in a polypeptide (the Tn antigen). Evidence suggests that this lectin also may require specific amino acid sequences at the receptor site of glycosylation. The disaccharide galactosyl (α-1,3) N-acetylgalactosamine is also a potent inhibitor of this lectin.Fluorescein labeled Vicia villosa lectin has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated fluorochromes. The excitation maximum is at 495 nm and the emission maximum is at 515 nm.

Lotus tetragonolobus lectin (LTL) encompasses a family of closely related glycoproteins with similar specificities toward α-linked L-fucose-containing oligosaccharides. Although many of the binding properties of Lotus lectin are similar to those of Ulex europaeus lectin I (UEL I), the binding affinities and some specificities for oligosaccharides are significantly different between these fucose-specific lectins. This fluorescein-labeled LTL features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Griffonia simplicifolia lectin I (GSL I) is a family of glycoproteins with molecular weights of approximately 114 kDa. Two subunits ("A" and "B") have slightly different molecular weights and combine to form tetrameric structures, resulting in five isolectins. The "A"-rich lectin preferentially agglutinates blood group A erythrocytes and appears to be specific for α-N-acetylgalactosamine residues, while the "B"-rich lectin preferentially agglutinates blood group B cells and is specific for α-galactose residues. Our GSL I is a mixture of the five isolectins and binds several glycoproteins, including laminin. This fluorescein-labeled GSL I features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Phaseolus vulgaris agglutinin (PHA) is a family of lectins, each consisting of four subunits. There are two different types of subunits: the "E" subunit (for erythroagglutinin) is involved primarily in red cell agglutination, and the "L" subunit (for lymphocyte) is involved in lymphocyte agglutination and mitogenic activity. These subunits combine to produce five isolectins. PHA-L has four "L" type subunits, does not agglutinate red cells, but is a potent mitogen. It is a specific marker for use in anterograde neuronal tracing. This fluorescein-labeled PHA-L features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is 100 mM acetic acid.

Lens culinaris agglutinin (LCA) contains two subunits of 17 kDa and two of 8 kDa. It has a narrower specificity than ConA because it recognizes sequences containing α-linked mannose residues and other sugars. An α-linked fucose attached to the N-acetylchitobiose of the core oligosaccharide enhances affinity. The narrower specificity allows subfractionation of glycoproteins and peptides after initial isolation with Con A. LCA prevents skin allograft rejection and can purify numerous glycoproteins, including immunoglobulins, histocompatibility antigens, and α2-macroglobulin. This conjugate of LCA to Dylight 649 features a ratio of fluorophore to lectin protein that provides optimal staining (excitation 655 nm, emission 670 nm). Supplied as a solution essentially free of unconjugated fluorophore, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is a mixture of 200 mM α-methylmannoside/200 mM α-methylglucoside.

Lycopersicon esculentum lectin (LEL or tomato lectin, TL) is a single-subunit glycoprotein containing arabinose and galactose and may form multimeric aggregates in solution. Although it shares specificities with potato lectin, Datura lectin, and wheat germ agglutinin, LEL is dissimilar in many respects. It binds well to glycophorin and Tamm-Horsfall glycoprotein, has been used to label vascular endothelium in rodents, and is an effective marker of blood vessels and microglial cells in rodents. Conjugation with a fluorophore facilitates fast, one-step detection and visualization using intravascular perfusion methods or direct application to tissue sections. This fluorescein-labeled LEL features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Sambucus nigra lectin (SNL) is isolated from elderberry bark and binds preferentially to sialic acid attached to terminal galactose in a-2,6 and to a lesser degree, a-2,3 linkage. Also called SNA (Sambucus nigra agglutinin) and EBL (elderberry lectin), SNL can inhibit cell-free protein synthesis. Binding is also inhibited to some extent by lactose or galactose. This lectin does not appear to bind sialic acid linked to N-acetylgalactosamine. SNA has been reported to inhibit cell-free protein synthesis. This fluorescein-labeled SNL features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is 500 mM lactose in buffered saline followed by 500 mM lactose in acetic acid.
